productImmunoDOT autoimmune panels are single-patient tests that use an enzyme immunoassay DOT technique for confirmation of specific antibodies and SS-A, SS-B, RNP, Sm, and DNA. A single-patient assay strip is incubated with dilute patient serum, allowing patient antibodies reactive with test antigens to bind to the solid support membrane. Only 10 µL of patient serum is needed for testing with each multiparameter assay strip. The reaction is enhanced by the removal of nonspecifically bound material, and alkaline phosphatase conjugated antihuman antibodies react with bound patient antibodies. Finally, the strip is transferred to an enzyme substrate reagent, which reacts with bound enzyme to produce an easily seen distinct dot.
