This company has developed an application for the determination of amylase on Hitachi analyzers. The method is based on the hydrolysis of 2-chloro-p-nitrophenyl-a-D-maltotrioside (CNPG3) by a-amylase, which release chloro-nitrophenol and forms 2-chloro-p-nitrophenyl-a-D-maltoside (CNPG2), maltotriose and glucose. The rate of increase in absorbance is measured at 405nm and is proportional to the a-amylase activity in the sample.
The ready-to-use liquid reagent has a 24-month shelf life when stored at 2-8ºC. It is linear to 2000 U/L.
