OptiBlaze™ ELISA femto-HRP, from G-Biosciences, St Louis, is a highly sensitive chemiluminescent detection system for horseradish peroxidase (HRP)-conjugated antibodies. The chemiluminescent substrates are ultrasensitive and stable, specifically developed for luminometer-based applications. It is able to detect low femtogram to picogram levels of enzyme, making it ideal for ELISA applications. The kit components contain premixed solutions and are enough for performing 1,000 reactions as per the protocol. The detection system is also available for the detection of alkaline phosphatase-conjugated antibodies during ELISA.

The Seeplex® Sepsis Test from Seegene, Rockville, Md, is a single lab test that quickly determines the levels of suspected targets in a patient’s blood sample to better inform the diagnosis and create an opportunity for quickly applying an appropriate treatment to head off the disease. The Seeplex Sepsis Test simultaneously screens for 64 sepsis-causing pathogens: 48 Gram (+) bacteria, 10 Gram (-) bacteria, and six fungi. Additionally, four drug-resistant genes (vanA, vanB, mecA, and blaSHV) can also be discriminated. The Seeplex Sepsis Test requires only 0.4 mL of a patient’s whole blood and provides test results in 5 hours. Seeplex Sepsis Test is not for diagnostic procedures in the United States and will not be available in the United States pending regulatory approval.

XIT Genomic DNA Kits
The XIT™ Genomic DNA kits from G-Biosciences, St Louis, allows isolation of DNA from a variety of sample types, including animal tissues, whole blood, buffy coat, bone marrow, gram positive and negative bacteria, plant tissues, mouse tail, yeast, and formalin-fixed, paraffin-embedded tissue. The kits use the principle of cell lysis, protein digestion and precipitation, and finally, DNA precipitation, to isolate high-quality genomic DNA. The purified DNA has an A260/A280 ratio from 1.7 to 2.0, and has high yields suitable for all types of downstream analyses.

This new technology provides test results within minutes, with enhanced sensitivity and detection comparable to laboratory-based oral fluids tests, and is offered in a six-panel format, which can simultaneously test for six drugs in each device, including THC (marijuana), cocaine, amphetamines, methamphetamines, phencyclidine, opiates, benzodiazepines, and/or methadone. Millipore Corp, Billerica, Mass, introduces its OxyELISA Oxidized Protein Quantitation kit, which provides all the reagents necessary for quantitative, high-throughput detection of modified proteins in cells undergoing oxidative stress. Oxidative stress is a cellular condition particularly relevant to aging and age-related diseases, such as Alzheimer’s disease, Parkinson’s disease, and dementia. The kit, by providing a fast and sensitive assay for oxidation, helps researchers, especially neuroscientists, understand the mechanism of cellular aging. As a result of oxidative stress, oxygen free radicals and other reactive species introduce carbonyl groups into proteins in the cell. The kit detects these carbonyl groups, which are hallmarks of the cellular oxidative stress level. The kit uses a direct labeled monoclonal antibody that is readily detected without a secondary antibody, making the ELISA assay more specific, easier to use, and faster. The OxyELISA kit complements Millipore’s more qualitative OxyBLOT™ assay, which detects oxidation of individual proteins after separation on a gel.

The GET™ Plasmid DNA Kits, by G-Biosciences, St Louis, are spin column purification kits for the isolation of high-quality plasmid from bacterial cultures. Three different formats of the kits are available—maxi, medi, or mini—which allow the isolation of various yields of plasmid. The kits use an extraction method based on alkaline lysis of bacteria and use high-affinity columns to bind and purify plasmid DNA. This quick and easy protocol eliminates toxic phenol/chloroform extractions or ethanol precipitations, and the extracted plasmid is then ready for further DNA manipulation, such as restriction enzyme digestion, ligation, transformations, transfections, and sequencing.
The VAP cholesterol test from Atherotech Inc, Birmingham, Ala, helps physicians pinpoint effective risk-lowering treatments for people with a family history or an existing condition of diabetes, high blood pressure, or heart disease—or who are already taking cholesterol-lowering medication. The cholesterol profile complies with updated National Cholesterol Education Program ATP III recommendations for LDL measurement, and the advanced lipid profile routinely reports all three lipoprotein parameters (LDL, non-HDL, and apoB), considered necessary by the American Diabetes Association and American College of Cardiology expert-consensus guidelines. The test directly measures LDL, which doesn’t require fasting for accurate measurement. The Diabetes and Combined Lipid Therapy Regimen study used the VAP Test to measure the effects of simvastatin alone, fenofibrate alone, and the combination of both on lipid subparticles in 300 patients (randomized) with Type 2 diabetes without existing heart disease. Researchers found that combining statin therapy with a fibrate is an effective way to correct an adverse cardiovascular risk profile in individuals with diabetes and mixed dyslipidemia (elevated LDL cholesterol and triglyceride levels combined with decreased levels of HDL cholesterol). The results of the 12-week, single-center, double-blind, placebo-controlled study were recently published in the American Journal of Cardiology.
Extraction buffer for solubilization of proteins
2D-Xtract™ protein solubilization/extraction buffer for 2D electrophoresis from G-Biosciences, St Louis, is a dry buffer that has optimized concentrations of urea, thiourea, CHAPS, and nondetergent sulfobetaine (ND SB) 201. Users add an appropriate volume of the supplied rehydration buffer to the dry buffer, and then use the mixture to solubilize proteins. The product is also designed to be used as a rehydration buffer for IPG strips. According to the company, 2D-Xtract minimizes the risk of modifying the native charge of the solubilized proteins due to carbamylation.
